Product Specific Information

The 4RA10 monoclonal antibody reacts with mouse CD162 (P-selectin glycoprotein ligand-1, PSGL-1).
This clone is validated for flow cytometry and can block CD162 binding to CD62P and CD62L.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.06 µg/test (100 µL final volume).
Cell number may range from 10⁵–10⁸ cells/test. Titrate antibody for optimal performance.

Fluorophore Information:
PerCP-eFluor 710 emits at 710 nm and is excited with the blue laser (488 nm).
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility.

Light Sensitivity: Tandem dye is sensitive to light; protect vial and samples from exposure.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-57)
  Performance after fixation should be verified empirically.

Target Information

CD162 (PSGL-1) is a 120 kDa homodimeric protein expressed on leukocytes and platelets.
It binds CD62P (P-selectin), CD62L (L-selectin), and CD62E (E-selectin) to mediate leukocyte adhesion, rolling, and extravasation.
CD162 serves as an immune marker of T cell exhaustion in chronic viral infection and cancer.
