Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (32–34 kDa), expressed as either CD8αβ heterodimer or CD8αα homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds MHC class I and associates with p56lck, playing a key role in T-cell development and activation.

Preliminary testing shows OKT8 does not compete with RPA-T8 or HIT8a clones for binding to human leukocytes, suggesting distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells at 5 µL (0.5 µg)/test (final volume 100 µL; 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL cell suspension.
• NovaFluor conjugates utilize Phiton™ technology for engineered fluorescence minimizing spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family. It acts as a co-receptor with the T cell receptor (TCR) for MHC class I molecules, facilitating T cell activation and signal transduction. CD8 exists as a CD8αβ heterodimer or CD8αα homodimer, expressed on T cells, NK cells, monocytes, and dendritic cells. Activation of CD8+ cytotoxic T lymphocytes (CTLs) is crucial for pathogen and tumor cell clearance, enhanced by IL-2, IL-12, and TGF-β1.
