Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule (32–34 kDa), expressed on thymocytes, mature T cells, and NK cells. CD8a binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

This antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. A test is defined as the amount of antibody that stains a sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Always add 5 µL of CellBlox Plus Blocking Buffer per sample for optimal staining.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in T cell signal transduction, differentiation, and activation. It functions as a co-receptor with TCR for MHC class I molecules. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells, existing as CD8αβ heterodimers or CD8αα homodimers. Upon MHC-I/peptide binding, it recruits Lck kinase to induce lymphokine production and CTL activation, essential for pathogen and tumor cell clearance.
