Product Specific Information

The HB5 monoclonal antibody reacts with human CD21, an approximately 145 kDa type I transmembrane protein expressed by mature B cells, follicular dendritic cells, and a subset of epithelial cells. CD21 acts as a receptor for complement component C3d and Epstein-Barr virus (EBV). It forms a complex with CD19 and CD81, contributing to B cell activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.02 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology, offering engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

CD21 (Complement Receptor 2, CR2, C3D receptor, EBV receptor) binds C3 complement fragments and plays a key role in immunocomplex uptake, memory B cell survival, and humoral immune response development. It also serves as a receptor for EBV and prions, facilitating neuroinvasion. CD21 interacts with CD23, CD19, CD81, and Leu13 to form a signal-transduction complex involved in B cell activation. Genetic variations in CD21 are linked to systemic lupus erythematosus type 9 (SLEB9). Multiple isoforms exist due to alternative splicing.
