Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. PD-L2, along with PD-L1, acts as a ligand for programmed death protein 1 (PD-1).
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma.

PD-L2 binds PD-1 with 2–6 times higher affinity than PD-L1, functioning as an inhibitory immune checkpoint molecule that regulates T cell activation and cytokine secretion (e.g., IFN-gamma, IL-10).
Interestingly, PD-L2 can also stimulate T cell proliferation even in PD-1-deficient cells, suggesting additional receptor interactions. Recent studies show PD-L2 also binds to RGMb (repulsive guidance molecule b), influencing respiratory tolerance through BMP and neogenin signaling pathways.

The human PD-L2 monomer comprises:
