Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 Alzheimer’s disease 환자의 뇌 아밀로이드 플라크의 주요 구성 성분입니다. Abeta peptide는 40~43개의 아미노산으로 이루어지며, beta-amyloid precursor protein (beta APP)으로부터 두 단계의 절단 과정을 통해 생성됩니다.
첫 번째 절단은 beta-secretase (BACE)에 의해 수행되며, 이후 gamma-secretase에 의해 추가 절단되어 amyloid beta peptide가 생성됩니다. Gamma-secretase는 presenilin-1, presenilin-2, nicastrin 등의 단백질 복합체로 구성되어 있습니다.
Abeta는 신경 섬유 엉킴(neurofibrillary tangles) 및 아밀로이드 코어의 주요 구성 요소이며, Alzheimer’s disease 외에도 Lewy body dementia, Down’s syndrome, cerebral amyloid angiopathy 등 다양한 신경퇴행성 질환에서 발견됩니다.
