Product Specific Information

The FJK-16s antibody reacts with Foxp3 from mouse, rat, dog, porcine, bovine, and cat. Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2) is a transcriptional regulator identified as the gene defective in scurfy mice. High expression of Foxp3 mRNA is observed in CD4+CD25+ regulatory T cells (Treg), and ectopic expression induces Treg phenotype.

Immunoblotting with FJK-16s maps the epitope to amino acids 75–125 of mouse Foxp3. This region is alternatively spliced in human but not in mouse splenocytes.
Optimized for use with Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00); other buffers are not recommended.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Mouse splenocytes using Foxp3/Transcription Factor Staining Buffer Set.
Recommended ≤ 1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).

Brilliant Violet™ 605 (BV605) emits at 605 nm and is compatible with violet (405 nm) laser. Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions.

  • Light Sensitivity: Protect from light.
  • Fixation: Compatible with IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54); store up to 3 days at 4°C in dark.
  • Not compatible with methanol-based fixation.
  • Excitation: 407 nm; Emission: 605 nm; Laser: Violet Laser.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis.

  • Expressed in CD25+CD4+ regulatory T cells, low in CD4+/CD25– cells, absent in CD4–/CD8+ T cells.
  • Defects cause scurfy phenotype in mice and IPEX syndrome in humans.
  • Regulated by p53 in response to DNA damage in human cancer cells.
