Description:
The monoclonal antibody 10C2 recognizes human nestin (residues 1464–1614). Nestin is a 220–240 kDa Class VI intermediate filament protein expressed in stem cells of the developing nervous system. Expression is also found in pancreatic islet and mesenchymal precursors, as well as cells with angiogenic potential. Upon differentiation, nestin-containing filaments are replaced by cell type-specific intermediate filament proteins such as GFAP. Expression has been identified in various cancer cells including glioblastomas and pancreatic tumors. No cross-reactivity to rodent nestins has been observed.

Applications Reported:
Useful for immunohistochemical staining of frozen and paraffin-embedded tissue sections, microscopy, and immunocytochemistry.

Applications Tested:
Validated for immunocytochemistry of formaldehyde-fixed and permeabilized human cells. Recommended usage ≤ 1 µg/mL, with titration for optimal assay performance.

Filter Recommendation:
For eFluor® 570 conjugates, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor® 555 or TRITC filters are also acceptable.

Target Information

Nestin is a Class VI intermediate filament abundantly produced in the developing central nervous system (CNS) and early embryonic neuroepithelial stem cells. It serves as a marker for stem/progenitor cells, glioma cells, and tumor endothelial cells, and is a superior angiogenic marker for evaluating tumor neovascularity. Highly specific antibodies to human Nestin aid in characterizing progenitor cell differentiation, enhancing therapeutic potential of neural stem/progenitor cells for CNS disease treatment, and identifying neuroepithelial tumor cells. Nestin expression is also seen in pancreatic hematopoietic stem cell populations and proliferating endothelial cells in colorectal cancer tissue.
