Target Information
Arg3.1 (ARC) is a master regulator of synaptic plasticity that self-assembles into virion-like capsids encapsulating RNAs for intercellular transfer in the nervous system. ARC is released from neurons via extracellular vesicles, transferring ARC mRNA into new target cells where it undergoes activity-dependent translation.
ARC capsids are endocytosed and can deliver ARC mRNA into neuronal cytoplasm. It is essential for protein synthesis-dependent long-term potentiation (LTP), long-term depression (LTD), and long-term memory formation. ARC regulates synaptic plasticity by promoting endocytosis of AMPA receptors (AMPARs), maintaining neuronal homeostasis through synaptic scaling.
Additionally, ARC mediates synapse elimination in the developing cerebellum, accumulates at weaker synapses to prevent undesired enhancement, and may facilitate elimination of synaptic material. ARC is also implicated in visual cortex plasticity and amyloid-beta processing via PSEN1 interaction. Beyond neuronal roles, ARC is expressed in skin-migratory dendritic cells, regulating fast migration and T-cell activation.
