Target Information
CD40는 종양괴사인자/신경성장인자 수퍼패밀리의 단일 사슬 당단백질로, Hodgkin 병 관련 항원 CD30과 상동성을 지닙니다. B세포(형질세포 제외), 일부 상피세포, 암세포 및 림프성 수지상세포에서 발현됩니다.
CD40은 T세포 의존성 면역글로불린 클래스 전환, 메모리 B세포 발달, 생식 중심 형성 등 다양한 면역·염증 반응의 매개에 필수적입니다.
AKNA 전사인자가 CD40 발현을 조절하며, TNFR2 어댑터 단백질은 CD40과 상호작용하여 신호전달을 매개합니다.
CD40–리간드 상호작용은 아밀로이드-베타 유도 미세아교세포 활성화에 필요하며, 알츠하이머병 초기 병리 과정과 관련된 것으로 알려져 있습니다.
CD40의 두 가지 상이한 스플라이싱 변이체가 보고되었으며, CD40 기능 이상은 Type 3 Hyper-IgM 면역결핍증 및 CD40 리간드 결핍증과 관련이 있습니다.
