Target Information
USP7 (HAUSP)는 ubiquitin 특이적 프로테아제(deubiquitylating enzyme)로, 기질 단백질로부터 ubiquitin을 절단합니다. 일반적으로 ubiquitylation은 단백질의 안정성과 분해에 관여하며, HAUSP의 활성은 기질 단백질을 안정화시키는 역할을 합니다.
HAUSP는 종양 억제 단백질 p53의 E3 ubiquitin ligase인 Mdm2의 직접적인 길항제로 알려져 있습니다. 정상 상태에서 p53은 Mdm2 매개 ubiquitination과 분해로 낮은 수준을 유지하지만, HAUSP는 p53을 탈-ubiquitination하여 Mdm2에 의한 분해를 억제함으로써 세포 스트레스에 대한 종양 억제 기능을 수행할 수 있습니다.
또한 HAUSP는 Myc, E1A 등의 온코진에 의해 활성화된 p53 안정화 과정에도 관여하며, 이는 p19ARF 의존적 또는 비의존적 경로를 통해 이루어질 수 있습니다. 이러한 점에서 HAUSP는 종양 억제 및 세포 보호 경로의 핵심 인자로 작용할 가능성이 있습니다.
