Target Information

Guanylate kinase (GK) catalyzes the ATP-dependent phosphorylation of GMP into GDP and is essential for recycling GMP and indirectly cGMP. GK is a highly conserved monomeric protein (~200 amino acids) found across prokaryotes, lower eukaryotes, and vertebrates.
It shares structural similarity with Vaccinia virus protein A57R (SalG2R). Proteins containing DHR, SH3, and GK-like domains are collectively termed MAGUKs (membrane-associated guanylate kinase homologs), including:

• Synaptic proteins (SAP90/PSD-95, CHAPSYN-110/PSD-93, SAP97/DLG1, SAP102)

GK contains an ATP-binding site (P-loop) in the N-terminal section, which is not conserved in GK-like domains of MAGUKs but retains residues involved in MP binding.
