Target Information
Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the γ phosphate of ATP, onto an acceptor amino acid in a substrate protein. Through this mechanism, protein kinases mediate signal transduction in eukaryotic cells, regulating metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.
With over 500 gene products, the protein kinase family is one of the largest in eukaryotes. Based on sequence comparisons of their tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains, the family is classified into eight major groups. The AGC kinase group includes 63 kinases such as:
- Cyclic nucleotide-regulated protein kinases (PKA & PKG)
- Diacylglycerol/phospholipid-dependent protein kinase C (PKC)
- RAC/Akt protein kinases
- G protein-coupled receptor kinases (ARK)
- Ribosomal protein S6 kinases (RSK)
