Presenilin 1은 조기 발병형 알츠하이머병 감수성의 지표로 처음 확인된 단백질입니다. Presenilin 1은 PEN2, nicastrin, APH-1과 함께 막결합 아스파틸 프로테아제 복합체인 γ-secretase를 형성하며, 이는 지질 이중층 내에서 특정 단백질을 절단합니다. 이 절단은 세포 표면 수용체 신호전달 과정에서 중요한 단계이며, 알츠하이머병 병리의 핵심인 신경독성 아밀로이드 펩타이드 생성에 관여합니다. γ-secretase는 APP의 막내 영역을 절단하여 비독성 p3 또는 독성 Abeta 펩타이드를 생성하며, Abeta 축적이 알츠하이머병의 전구체로 알려져 있습니다. Presenilin 1은 여러 동형체가 존재합니다.
