Target Information
The expression of DUSP1 gene is induced in human skin fibroblasts by oxidative/heat stress and growth factors. It encodes a protein with structural features similar to non-receptor-type protein-tyrosine phosphatase family members and shows significant amino acid sequence similarity to a Tyr/Ser-protein phosphatase encoded by the vaccinia virus late gene H1. The bacterially expressed and purified DUSP1 protein exhibits intrinsic phosphatase activity, specifically inactivating MAP kinase in vitro by dephosphorylating both phosphothreonine and phosphotyrosine residues. It also suppresses MAP kinase activation by oncogenic Ras in Xenopus oocyte extracts. Therefore, DUSP1 is considered important in the human cellular response to environmental stress and in the negative regulation of cellular proliferation.
