Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), essential for DNA biosynthesis.
TS is a critical target for fluoropyrimidines, a major class of antineoplastic drugs used in solid tumor treatment.
In tumor cells, 5-FU and fluorodeoxyuridine are converted to FdUMP, which inactivates TS by forming a ternary covalent complex with the folate cofactor 5,10-methylenetetrahydrofolate.
TS protein expression correlates with response to 5-fluorouracil (5-FU) in colorectal, gastric, head and neck, and breast carcinomas.
