Target Information
Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), a critical step in DNA biosynthesis. TS is a key target for fluoropyrimidines, a class of antineoplastic drugs used to treat solid tumors. Both 5-fluorouracil (5-FU) and fluorodeoxyuridine are converted to FdUMP in tumor cells, forming a ternary covalent complex with TS and the folate cofactor 5,10-methylenetetrahydrofolate. TS expression correlates with response to 5-FU therapy in colorectal, gastric, head and neck, and breast carcinomas.
