Target Information

Progression through the cell cycle requires activation of a series of cyclin-dependent kinases (Cdks). The catalytic subunit Cdk2 is inactive alone and partially activated by binding regulatory cyclins such as cyclin D1, with full activation requiring phosphorylation at Thr160. The enzyme responsible for this phosphorylation, Cdk-activating kinase (CAK), comprises a catalytic subunit, a regulatory subunit (cyclin H), and a subunit of unknown function. The regulatory subunit Mat1 associates with the cyclin H–Cdk7 complex throughout the cell cycle, forming the active Cdk-cyclin complex. Cyclin H–Cdk7–Mat1 complexes also phosphorylate the carboxy terminus of RNA polymerase II, highlighting Mat1 as an essential assembly factor for Cdk-cyclin activation.
