Product Specific Information
The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. The epsilon chain, along with CD3 gamma and delta, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Crosslinking of TCR via immobilized UCHT1 induces intracellular signaling leading to activation and proliferation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.1 µg)/test in a final 100 µL volume. Suitable for 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability with NovaFluor dyes.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL buffer per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton technology based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimized spillover and spread.

Target Information
The CD3 complex transduces antigen-recognition signals into T-cell cytoplasm and regulates TCR surface expression. It consists of CD3γ, CD3δ, CD3ε, and CD3ζ subunits, structurally related members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with intracellular signaling molecules through tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. CD3 gene defects are associated with CD3 immunodeficiency.
