Target Information

The E3 ubiquitin-protein ligase NEDD4 is a Ubl conjugation pathway protein containing one HECT (E6AP-type E3 ubiquitin-protein ligase) domain and four WW domains.
It participates in the degradation pathway of VEGFR-2/KDFR, independent of ubiquitin-ligase activity, and functions as a potential proto-oncogene by negatively regulating PTEN via ubiquitination.
NEDD4 is also involved in viral budding and regulation of epithelial Na⁺ channels, influencing arterial hypertension.
It localizes to the sarcolemmal region of muscle fibers, and its increased expression is associated with muscle unloading, denervation, and decreased Notch1 expression, acting as a downregulator of Notch-1 gene expression.
