Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except erythrocytes and platelets. The cytoplasmic portion has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells. Recommended use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample to antibody cocktails prior to cell combination. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor Technology:
Based on Phiton™ technology using novel nucleic acid dye structures engineered for minimal spillover and spread.
Excitation: 494 nm; Emission: 585 nm; Laser: 488 nm (Blue).

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). Absent on non-hematopoietic cells. Plays critical roles in lymphocyte development, antigen signaling, and regulation of Src-family kinases. Isoforms (e.g., CD45RA) differ by alternative splicing and cell-type-specific expression. CD45 regulates T/B-cell receptor signaling, JAK kinase activity, integrin-mediated survival pathways, and apoptosis-related processes.
