Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. It plays a critical role in lymphocyte activation through its tyrosine phosphatase enzymatic activity.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample or per 100 µL of cell sample for single-color controls.
  • Add to antibody cocktails before combining with cells.

Target Information

CD45 (Leukocyte Common Antigen, LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, representing ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues and cell lines. CD45 regulates Src-family kinases, T- and B-cell receptor signaling, cytokine signaling, and integrin-mediated pathways, playing key roles in immune cell activation and survival. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
