Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets, and plays a key role in lymphocyte activation through its tyrosine phosphatase activity.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells
• Recommended use: 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, offering engineered fluorescent signatures with minimized spillover and spread effects.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, constituting ~10% of lymphocyte surface proteins. It is absent on non-hematopoietic cells and tissues. CD45 regulates Src-family kinases, plays a critical role in lymphocyte development, antigen receptor signaling, and cytokine receptor regulation. Isoforms (e.g., CD45RA) differ by alternative splicing and expression pattern across lymphoid subtypes. CD45 also modulates integrin-mediated signaling, apoptosis, and immune function regulation.
