ATG5 (Autophagy Related 5)는 자가포식(autophagy)에 필수적인 요소로, 세포 자멸(apoptosis) 과정에서도 중요한 역할을 합니다.
ATG5는 다음과 같은 다양한 세포 기능에 관여합니다:
- 산화 손상 후 미토콘드리아 품질 관리
- 선천적 항바이러스 면역 반응의 음성 조절
- 림프구 발달 및 증식
- MHC II 항원 제시
- 지방세포 분화 조절
ATG12와의 결합 후, autophagosome 형성에 참여하며, Fas-associated protein with death domain (FADD)와 상호작용하여 autophagic cell death를 유도합니다.
ATG5-ATG12 복합체는 autophagosome 형성 직전 또는 직후에 막에서 분리되는 컵 형태의 분리막을 형성합니다. 또한 RIG-I 및 VISA(IPS-1)와 같은 선천적 면역 단백질과 결합하여 type I interferon 생성을 억제하고 바이러스 복제를 허용합니다.
