Target Information
ATG5 (Autophagy Related 5)는 오토파지에 중요한 요소로, 세포 자가사멸(apoptosis) 과정에도 관여합니다. 또한 산화 손상 후 미토콘드리아 품질 관리, 선천적 항바이러스 면역 반응의 음성 조절, 림프구 발달 및 증식, MHC II 항원 제시, 지방세포 분화 등 다양한 세포 과정에 참여합니다.
ATG12와 결합 후 오토파고좀 형성에 관여하며, FADD와 상호작용하여 오토파지성 세포 사멸을 유도합니다. ATG5-ATG12 복합체는 오토파고좀 형성 직전 또는 직후에 막으로부터 분리되는 컵 모양의 격리막을 형성합니다.
또한 RIG-I 및 VISA(IPS-1)와 같은 선천 면역 반응 단백질과 결합하여 type I interferon 생성을 억제하고 바이러스 복제를 허용합니다. ATG5 기능 이상은 spinocerebellar ataxia 등 질환과 관련이 있습니다.
