Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a stem cell marker, with reversible expression on human hematopoietic stem cells. CD34 may function as a receptor mediating endocytosis and regulating adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors.

CD34 likely represents a specific developmental state of hematopoietic cells with distinct adhesion and differentiation properties. It may act as an adhesion molecule facilitating stem cell attachment to the bone marrow extracellular matrix or stromal cells. Additionally, CD34 could serve as a scaffold for lineage-specific glycans, enabling stem cell interactions with lectins from stromal or marrow components. Its intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
