Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 minutes.
Target Information
Estrogen Receptor beta (ER beta, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ER beta was initially cloned and characterized from testis.
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, induces a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA.
ER beta binds estrogens with an affinity similar to ER alpha. Some ER beta isoforms dominantly inhibit ER alpha activity in reproductive tissues. ER beta is widely expressed and may act as a tumor suppressor, implicated in multiple diseases such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain Disease, and cardiovascular disease risk.
