Target Information
TLR2 (Toll-like receptor 2) is a member of the TLR family that plays a fundamental role in pathogen recognition and activation of innate immunity. TLR2 acts through adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKC alpha/beta and NF-kappa-B.
TLR2 can form heterodimers with TLR1 or TLR6, enabling recognition of bacterial and mycoplasma lipoproteins. TLRs are conserved from Drosophila to humans and share structural and functional similarities.
TLR2 is highly expressed in peripheral blood leukocytes and mediates host responses to Gram-positive bacteria and yeast via NF-kappaB activation. It recognizes pathogen-associated molecular patterns (PAMPs) and promotes cytokine production for effective immunity.
TLR2 is expressed in human tonsils, microglia, lymph nodes, appendices, and activated B-cells. CD14+ monocytes show highest TLR2 expression, followed by CD15+ granulocytes, CD19+ B-cells, and CD3+ T-cells.
Expression is regulated by immune modifiers: LPS, GM-CSF, IL-1, and IL-10 upregulate TLR2; IL-4, IFN-gamma, and TNF downregulate expression in monocytes.
