Target Information

ZO-3 is a member of the membrane-associated guanylate kinase (MAGUK) family and functions as a tight junction-associated protein. Tight junctions form intercellular boundaries between epithelial and endothelial cells.

ZO-3 has both structural and signaling roles and contains PDZ, SH3, and guanylate kinase (GuK) domains. ZO-1 forms complexes with either ZO-2 or ZO-3, and these proteins associate with claudin, occludin, and F-actin at tight junction strands, linking the actin cytoskeleton to the junction.

ZO-1 expression is reduced in many breast cancer lines. ZO-2 and ZO-3 are ubiquitously expressed in epithelial tight junctions, while ZO-1 is also found in cardiac myocytes.
