Target Information
Phosphoinositide-specific phospholipase C (PLC)는 수용체 매개 신호전달의 시작에 중요한 역할을 하며, phosphatidylinositol 4,5-bisphosphate로부터 두 가지 2차 신호전달물질인 inositol 1,4,5-triphosphate와 diacylglycerol을 생성합니다.
PLC는 여러 이소형(PLCβ1, β2, β3, β4, γ1, γ2, δ1, δ2, ε)을 포함하며, PLCδ는 네 가지 isoform으로 존재합니다. PLCδ1은 칼슘 신호 증폭기이며, 비정형 GTP 결합 단백질에 의해 활성화됩니다. 또한 PLCδ1은 transglutaminase II 매개 옥시토신 수용체 및 α1B-아드레노수용체 신호전달의 효과기 역할을 합니다.
Mouse PLCδ1은 뇌, 심장, 폐, 고환에서 높은 발현을 보이며, Alzheimer’s disease(AD) 환자 뇌에서 비정상적으로 축적되어 병리적 역할을 할 가능성이 있습니다. PLCδ2는 위암 및 장상피 화생에서 특이적으로 발현되며, 위암성 변환의 잠재적 마커로 고려됩니다.
