For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

Tuberous sclerosis complex (TSC) is an autosomal dominant tumor syndrome caused by mutations in either TSC1 or TSC2 tumor suppressor genes.
These gene products form a complex that indirectly reduces mammalian Target of Rapamycin (TOR) signaling, which regulates cell growth and cycle via nutrient and growth factor integration.
TOR activity is stimulated by Rheb, a Ras superfamily G-protein, when its bound GTP/GDP ratio is high.
Immunoprecipitated TSC1/TSC2 stimulates Rheb GTPase activity in vitro, reducing Rheb’s ability to activate TOR.
Overexpression of TSC1 and TSC2 decreases the GTP/GDP ratio bound to Rheb, inhibiting cell growth.
A shorter 40 kDa isoform of TSC1 exists, but its function remains unknown.
