Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains, recognizing most of the TCR V alpha 2 subfamily in mice with a, b, and c haplotypes. It also reacts with V delta 8 products due to high sequence homology.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse lymph node cells; ≤0.5 µg/test (100 µL final volume). Recommended preblocking Fc receptors with purified anti-mouse CD16/32 (Product #14-0161-85).
• Optimization: Antibody should be titrated for optimal performance.

Super Bright 780 Dye Characteristics

• Tandem dye excited by violet laser (405 nm), emission at 780 nm.
• Recommended bandpass filter: 780/60.
• Use Super Bright Complete Staining Buffer (Product #SB-4401) when using multiple Super Bright dyes to minimize non-specific interactions.
• Compensation values may differ between bead-based and cell-based setups; compensation with cells is recommended.
• Sensitive to photo-induced oxidation; protect from light.
• Samples can be fixed using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

T cell receptors (TCR) discriminate foreign from self-peptides presented by MHC class II molecules, crucial for adaptive immunity. Recognition of self-peptides relates to autoimmune diseases and tumor-associated peptides. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to V beta segments are found in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially moderating inflammatory responses.
Most human T cells express TCR alpha-beta with CD4 or CD8. Double negative (DN) T cells lacking both markers increase in autoimmune or immunodeficiency disorders. Gamma-delta T cells, found mainly in epithelia, exhibit limited diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory activity.
