Target Information
Functions as a key mediator in apoptosis and inflammation. Promotes caspase-mediated apoptosis involving predominantly caspase-8 and also caspase-9 in a probable cell type-specific manner. Involved in activation of the mitochondrial apoptotic pathway and promotes caspase-8-dependent proteolytic maturation of BID independently of FADD in certain cell types. Mediates mitochondrial translocation of BAX and activates BAX-dependent apoptosis coupled to activation of caspase-9, -2, and -3.
In macrophages, participates in pyroptosis, a caspase-1-dependent inflammatory cell death, and is a major component of the ASC pyroptosome that recruits and activates caspase-1. Acts as an adapter in the assembly of inflammasomes that activate caspase-1, leading to secretion of proinflammatory cytokines. Required for recruitment of caspase-1 to inflammasomes containing pattern recognition receptors such as NLRP2, NLRP3, AIM2, and possibly IFI16. Facilitates procaspase-1 processing in NLRP1 and NLRC4 inflammasomes.
Also involved in inflammasome activation triggered by bacterial muramyl dipeptide (via NOD2), and may participate in DDX58-triggered proinflammatory responses. Isoform 2 may regulate inflammasome adapter activity, while isoform 3 inhibits IL-1β maturation. In collaboration with AIM2, may mediate caspase-8-dependent cell death independent of caspase-1. In adaptive immunity, contributes to dendritic cell maturation and T-cell activation, and may regulate DOCK2 expression and NF-κB signaling.
