Product Specific Information
The eBioSN4 monoclonal antibody reacts with human CD9, a 24 kDa tetraspanin family protein with four hydrophobic transmembrane domains. CD9 is expressed in platelets, eosinophils, basophils, pre-B cells, activated T cells, and neural cell lines. Its expression is associated with malignant phenotypes, including 90% of non-T cell acute lymphoblastic leukemia and 50% of chronic lymphocytic and acute myeloblastic leukemias.
In platelets, CD9 is expressed in alpha-granules and associates with integrin αIIb/βIII to mediate platelet aggregation. CD9 also induces pre-B cell aggregation, adhesion, and migration of Schwann cells, and provides co-stimulatory signals for T cells independently of CD28.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human platelets at 5 µL (0.25 µg) per test. A test corresponds to staining in 100 µL final volume with 10⁵–10⁸ cells/test.
Fluorophore Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter and ensure instrument compatibility.
Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Light Sensitivity:
Protect from light to prevent photo-induced oxidation.
Fixation:
Samples can be stored using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information
CD9 antigen is a surface glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells. It belongs to the tetraspanin family, spanning the membrane four times.
In pre-B cells and platelets, CD9 regulates activation and aggregation via integrin CD41/CD61 (GPIIb/GPIIIa). CD9 is involved in cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, and sperm-egg fusion. It contributes to cell attachment, proliferation, and effective T cell stimulation through MHC II association on dendritic cells.
CD9 plays roles in differentiation, adhesion, signal transduction, and suppression of cancer cell motility and metastasis, acting as a metastasis suppressor in solid tumors.
