Target Information
Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two subclasses: NAD(+)-dependent and NADP(+)-dependent.
Five isocitrate dehydrogenases have been reported:
- Three NAD(+)-dependent isocitrate dehydrogenases (mitochondrial matrix)
- Two NADP(+)-dependent isocitrate dehydrogenases (one mitochondrial, one cytosolic)
The NADP(+)-dependent cytoplasmic isozyme is a homodimer and is found in the cytoplasm and peroxisomes. It contains the PTS-1 peroxisomal targeting signal sequence, suggesting roles in NADPH regeneration for intraperoxisomal reductions and reactions involving 2-oxoglutarate, such as alpha-hydroxylation of phytanic acid.
The cytoplasmic enzyme plays a key role in cytoplasmic NADPH production.
