Product Specific Information

Description
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens are associated with beta 2-microglobulin and expressed by all nucleated human cells, playing a central role in immune response and tumor surveillance. The antibody recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity has been observed in baboon, rhesus, and cynomolgus monkey.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test in 100 µL final volume, with 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining with NovaFluor conjugates.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

HLA-class I MHC antigens are intrinsic membrane glycoproteins expressed on nucleated cells and associated with beta2-microglobulin. They present foreign peptides for recognition by cytotoxic T cells, critical for antiviral and antitumor immunity.
HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding the identification and characterization of neoantigens using LC-MS/MS.
