Product Specific Information
The W6/32 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class I, HLA-A, B, C.
MHC class I antigens associated with beta 2-microglobulin are expressed by all human nucleated cells and play a central role in cell-mediated immune response and tumor surveillance.
W6/32 mAb recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Crossreactivity is seen in baboon, rhesus, and cynomolgus monkey.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.6 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
Add 5 µL per sample before combining antibody cocktails with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.

HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells and associated with beta2 microglobulin.
They present foreign determinants to cytotoxic T cells, critical for antiviral and antitumor defense.
Human HLA-class I antigens include HLA-A, HLA-B, and HLA-C molecules.
HLA and MHC antibodies are essential in Immunopeptidomics, enabling identification and characterization of neoantigens via LC-MS/MS.
