Product Specific Information

The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens, associated with beta 2-microglobulin, are expressed by all nucleated human cells and are central to cell-mediated immune response and tumor surveillance.
W6/32 mAb recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates 43 kDa and 11–12 kDa chains. Crossreactivity observed in baboon, rhesus, and cynomolgus monkey.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (1 µg) per test in a 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining antibodies with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

HLA-class I MHC antigens are intrinsic membrane glycoproteins expressed on nucleated cells, associated with beta2 microglobulin. They mediate immune recognition by cytotoxic T cells, playing a key role in antiviral and antitumor defense.
HLA and MHC antibodies are essential in immunopeptidomics for identifying and characterizing neoantigens via LC-MS/MS analysis.
