Target Information
Modification of target proteins by ubiquitin participates in various biological functions. Proteins destined for degradation or processing via the 26S proteasome are coupled to multiple copies of ubiquitin. Attachment of ubiquitin or ubiquitin-related molecules may alter subcellular distribution or protein activity. Deubiquitination, catalyzed by deubiquitinating enzymes, adds another layer of regulation. These enzymes fall into four families:
- Ubiquitin C-terminal hydrolases
- Ubiquitin-specific processing proteases (USPs)
- OTU-domain ubiquitin-aldehyde-binding proteins
- Jab1/Pad1/MPN-domain-containing metallo-enzymes
Among these, USPs are the most widespread deubiquitinating enzymes across evolution. They release ubiquitin from conjugated proteins and contain conserved Cys and His boxes, with variable N- and C-terminal extensions that contribute to substrate recognition, localization, and protein interactions.
