STAT3는 STAT (Signal Transducers and Activators of Transcription) 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동하여 전사인자로 작용합니다. STAT3는 CREB-binding protein과 같은 코활성화 인자가 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화됩니다. Tyrosine 705의 인산화는 STAT3의 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다. 인간에서는 STAT3 유전자가 17번 염색체 q arm에 위치하며, 다양한 세포 성장 및 세포사 과정에 핵심적인 역할을 합니다. Rac1, PIAS3 등의 단백질이 STAT3의 활성 조절에 관여하며, 세 가지 스플라이싱 변이체가 보고되었습니다. STAT3 유전자 결손 마우스는 배아 초기 단계에서 치명적입니다.
