Target Information

NUAK1 (ARK5) is a serine/threonine kinase involved in various cellular processes. Overexpression has been associated with tumor invasion and metastasis, particularly in colorectal cancer.
It regulates cellular senescence and ploidy by mediating phosphorylation of Ser-464 on LATS1, controlling its stability.
ARK5 influences cell adhesion through regulation of the myosin protein phosphatase 1 (PP1) complex by phosphorylating PPP1R12A, which interacts with 14-3-3, reducing dephosphorylation of myosin MLC2.
It may participate in DNA damage response by phosphorylating p53/TP53 at Ser-15 and Ser-392, and is recruited to the CDKN1A/WAF1 promoter to aid transcription activation.
ARK5 can promote tumor invasion and metastasis under AKT1 regulation and phosphorylation. It suppresses Fas-induced apoptosis by phosphorylating CASP6, preventing caspase activation and CFLAR cleavage.
Additionally, it regulates UV-induced DNA damage response via CDKN1A phosphorylation in association with STK11, contributing to CDKN1A degradation necessary for DNA repair.
