Product Specific Information
The 235D monoclonal antibody recognizes human SPI-B, a member of the ETS transcription factor family involved in lymphoid development and activity. SPI-B binds the PU box (5-GAGGAA-3) and is expressed in mature B cells, immature T cells, plasmacytoid dendritic cells, and intestinal microfold (M) cells. In human tonsil tissue, high SPI-B expression is observed in plasmacytoid dendritic cells and lower levels in germinal center B cells.

This antibody is suitable for western blotting, immunohistochemistry (FFPE), microscopy, and immunocytochemistry.
Applications Tested:
Validated for IHC on formalin-fixed paraffin-embedded human tissue using low pH antigen retrieval. Recommended concentration ≤5 µg/mL. Optimal dilution should be determined experimentally.
Target Information
Spi-B is a transcription factor belonging to the ETS family, characterized by a conserved ETS domain for specific DNA binding. It interacts with IRF4, CREBBP, EP300, SPI1/PU.1, JUN, and TBP. Spi-B binds to the PU-box (5-GAGGAA-3), acting as a lymphoid-specific enhancer. Two isoforms are known. Spi-B is primarily expressed in plasmacytoid dendritic cells (pDCs), promoting their development and antiviral response. It is not expressed in T cells or granulocytes but may be present in liver stem cells and B cells, where it plays roles in BCR signaling and B-cell development.
