Product Specific Information

The monoclonal antibody 1F3 recognizes human TRAF-1, a multifunctional adapter molecule in the TNF receptor associated factor (TRAF) family. TRAF-1 regulates cell survival and apoptosis, mediates NF-κB and JNK signaling, and forms a heterodimeric complex with TRAF-2. It interacts with receptors such as CD30, CD40, CD357 (AITR), and cytoplasmic proteins including NIK, RIP2, A20, and TRIP. Expression of TRAF-1 can be induced by Epstein-Barr Virus (EBV) and TNF-α.

Applications Reported: Suitable for western blotting, microscopy, immunohistochemistry, and immunocytochemistry.
Applications Tested: Validated for IHC of formalin-fixed paraffin-embedded human tissue using low pH antigen retrieval at ≤5 µg/mL. Optimal dilution should be determined experimentally.

Target Information

The TRAF-1 protein is a member of the TNF receptor (TNFR) associated factor family, mediating signal transduction from TNFR superfamily receptors. TRAF-1 and TRAF-2 form a complex essential for TNF-α-mediated activation of MAPK8/JNK and NF-κB pathways. This complex also interacts with inhibitor-of-apoptosis proteins (IAPs), contributing to anti-apoptotic signaling. Expression can be induced by Epstein-Barr Virus (EBV), and the viral LMP1 protein interacts with TRAF proteins, linking EBV-mediated B cell transformation to TNFR signaling. Multiple transcript variants encoding different isoforms have been identified.
