Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as either CD8αβ heterodimer or CD8αα homodimer.
CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported
- Flow cytometric analysis of mouse splenocytes.
Applications Tested
- Tested by flow cytometry; use ≤ 0.5 µg/test (100 µL final volume).
- Cell number range: 10⁵–10⁸ cells/test.
- Antibody should be titrated for optimal performance.
Important Notes
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
- Add 5 µL per sample (regardless of number of NovaFluors).
- For single-color controls, use 5 µL per 100 µL cell sample.
NovaFluor Technology
NovaFluor conjugates are based on Phiton™ technology, using nucleic acid dye structures for engineered fluorescent signatures minimizing spillover and spread.

Note: Internal testing shows non-specific B cell staining in SJL mice; not observed in BALB/c or C57BL/6 mice.
Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein involved in signal transduction, T cell differentiation, and activation.
Expressed as a CD8αβ heterodimer on T cells and CD8αα homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells.
Functions as a co-receptor for MHC class I molecules, recruiting Lck kinase upon ligation, leading to CTL activation and pathogen/tumor clearance.
Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
