Target Information
PREP-2 (Pbx-regulating protein-2), also known as PBX/knotted 1 homeobox 2 or PKNOX2, is a DNA-binding protein belonging to the TALE (three amino acid loop extension)/MEIS family. It forms stable complexes with Pbx proteins and is homologous to PREP-1, though with a more restricted tissue distribution and higher DNA-dissociation rate. PREP-2 forms a heterodimer with Pbx1, which relocates to the nucleus to associate with HoxB1, forming a ternary complex. Unlike PREP-1, which increases transcriptional activation, PREP-2 slightly decreases transcriptional activity. Multiple isoforms exist, localized in the nucleus or cytoplasm, with cytoplasmic forms co-localizing with F-actin, G-actin, and tubulin/microtubules.
