Target Information

DR5 (Apo2, TRAIL-R2, TRICK2, KILLER) is a death domain-containing receptor for TRAIL, mediating TRAIL-induced apoptosis. It belongs to the TNF-receptor superfamily and contains an intracellular death domain. DR5 activation by TNFSF10/TRAIL/APO-2L transduces an apoptosis signal. FADD-deficient mouse studies indicate that FADD is required for DR5-mediated apoptosis. Two transcript variants encoding different isoforms and one non-coding transcript exist. Overexpression induces apoptosis and activates NF-kappa-B. DR5 is expressed in various cell types, with high levels in lymphocytes and spleen. DR5 dysfunction is associated with squamous cell carcinoma and diffuse infiltrative lymphocytosis syndrome.
