Product Specific Information
This RR3-16 monoclonal antibody reacts with the mouse T cell receptor (TCR) V alpha 3.2 chain. The TCR is composed of alpha and beta chains, and specificity is determined by Va, Ja, Vb, Db, and Jb gene rearrangement.
The RR3-16 antibody recognizes the V alpha 3.2 chain on T cells from mouse strains bearing the b (e.g., C57BL/6) or c haplotype (e.g., SJL, SWR, NOD) in the Tcra gene complex.
The V alpha 3.2 chain is absent in mice with the a (e.g., Balb/c, AKR, C3H) and d (e.g., DBA/1, DBA/2) haplotypes.
It is more highly expressed on CD8+ T cells.
- Applications Reported: Flow cytometric analysis
- Applications Tested: Tested on mouse lymph node cells. Use ≤ 0.25 µg/test (defined as staining a cell sample in 100 µL final volume).
Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best performance. - Excitation: 633–647 nm
- Emission: 660 nm
- Laser: Red Laser
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
T cell receptors (TCRs) distinguish foreign from self-peptides presented by MHC class II molecules, essential for adaptive immune response.
Self-peptide recognition is linked to autoimmune disease, while mutant self-peptides can be tumor-associated.
Superantigen engagement of TCRs can cause toxic shock syndrome.
Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially modulating TH1-mediated inflammation.
Most human T cells express alpha-beta TCRs along with CD4 or CD8.
A small subset lacks both (double negative, DN), often elevated in autoimmune or immunodeficiency disorders.
Gamma-delta T cells, found mainly in epithelia, show less diversity and distinct antigen recognition, with some subsets exhibiting antitumor and immunoregulatory activity.
