Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, which are involved in G1 or G2/M arrest.
Structurally, p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 has been associated with tumor formation. p53 functions to block proliferation of damaged cells and acts as an anticancer agent. Mutations in the TP53 gene are found in many human cancers, leading to loss of tumor suppressor activity. Germline mutations are also associated with cancer-prone conditions such as Li-Fraumeni syndrome.
