Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 activates transcription of genes such as p21waf1, contributing to G1 or G2/M arrest.

The structure of p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 functions as a key anticancer agent by preventing proliferation of damaged cells. It is also linked to aging, oxidative stress, and cognitive functions such as long-term potentiation (LTP).

p53 is expressed at low levels in normal cells but highly in transformed cell lines, contributing to malignancy. Mutations in p53 disrupt DNA binding and tumor suppressor activity, and TP53 gene alterations occur both somatically and germline, notably in Li-Fraumeni syndrome.
