Gamma-glutamylcysteine synthetase (gamma-GCS) is the rate-limiting enzyme for glutathione (L-gamma-glutamyl-L-cysteinylglycine, GSH) synthesis. GSH is a vital antioxidant present in mammalian cells, providing intra- and extracellular protection. Gamma-GCS is a heterodimer consisting of a heavy catalytic subunit and a light regulatory subunit, responsive to inflammation, phenolic antioxidants, heat shock, oxidants, and cytokines.
